Аннотация: Tuberculosis (TB) is a serious world health problem. Traditional methods using for the study of Mycobacterium tuberculosis (Mtb) do not provide information about the amount and functional state of Mtb, especially if Mtb are located in alveolar macrophages. This knowledge about Mtb is very important for the selection of the best treПоказать полностьюatment for TB patients.??The aim is to create the method for the production of ex vivo cultures of alveolar macrophages produced from lung specimens of TB patients.??Lung tissue samples were obtained after surgery of TB patients. The cells obtained from lung pieces were placed in RPMI 1640 medium with fetal bovine serum to plastic plates with cover glasses in the bottom and cultured for 16–18 h. Then, the cells on cover glasses were fixed and stained by Ziehl-Nelsen method or with the use of antibodies and other reagents.??We obtained the ex vivo cell cultures largely composed by alveolar macrophages (90-99%) with and without Mtb. Lymphocytes, fibroblasts, neutrophils, and multinucleate Langhans giant cells were observed too. Alveolar macrophages contained a single Mtb or Mtb in colonies, including with cord morphology of Mtb growth. All macrophages were stained on CD14 receptor to bacterial lipopolysaccharides as the major macrophage/monocyte-specific marker. The Nile red-positive foamy alveolar macrophages with and without Mtb were determined in the ex vivo cell cultures.??For the first time, we studied the number of cells with Mtb and functional state of Mtb in host cells in the lungs of TB patients with use of the new method created by us for correction of drug regimen of TB patients after surgery and estimation of their epidemiological danger.
